چکیده
The RECELL® Autologous Cell Harvesting Device enables point-of-care preparation of autologous skin cell suspension (ASCS). Currently, ASCS is approved for the treatment acute thermal burn injuries, with additional indications under investigation. We hypothesize therapeutic potential may be enhanced regeneration and primed rejuvenation by improving cells’ ability to divide differentiate reverse-aging through telomere extension. A method safely extend telomeres in fibroblasts myoblasts has been demonstrated using delivery modified mRNA encoding telomerase (TERT). This increased activity, delayed expression senescence markers, proliferative capacity, while avoiding immortalization insertional mutagenesis. Notably, our previous studies showed TERT therapy reversed vascular extended lifespan progeria mice. purpose this work demonstrate feasibility modifying activity lipid nanoparticles (LNPs). uptake downstream protein were evaluated vitro various LNPs. Data from Incucyte FACS experiments onset mRNA-LNP cationic LNPs 15-30min, yielding transfection efficiency up 95% within 12h. Telomerase was identified transfected a TRAP assay, significant increase observed over control. Animal an immunocompromised mouse transplanted human derived are ongoing understand impact on regeneration. Methods have developed efficiently transfect substantially activity. novel approach holds promise other RNA-based cellular therapies aimed at cutaneous defects.

Bilayer hydrogel with autologous stem cells derived from debrided human burn skin for improved skin regeneration.
The objective of this study was to demonstrate that stem cells isolated from discarded skin obtained after debridement can be used with collagen and fibrin-based scaffolds to develop a tissue-engineered vascularized dermal equivalent. Discarded tissue samples were collected from severely burned patients undergoing wound debridement. Stem cells were isolated from the adipose tissue layer and the...
متن کاملA Novel Method for Skin Lesion Segmentation
Skin cancer has been the most usual and illustrates 50% of all new cancers detected each year. If they detected at an early stage, treatment can become simple and economically. Accurate skin lesion segmentation is important in automated early skin cancer detection and diagnosis systems. The aim of this study is to provide an effective approach to detect the skin lesion border on a purposed imag...

متن کاملStem Cell Niches for Skin Regeneration
Stem cell-based therapies offer tremendous potential for skin regeneration following injury and disease. Functional stem cell units have been described throughout all layers of human skin and the collective physical and chemical microenvironmental cues that enable this regenerative potential are known as the stem cell niche. Stem cells in the hair follicle bulge, interfollicular epidermis, derm...
متن کاملHealing Potential of Mesenchymal Stem Cells Cultured on a Collagen-Based Scaffold for Skin Regeneration
Background: Wound healing of burned skin remains a major goal in public health. Previous reports showed that the bone marrow stem cells were potent in keratinization and vascularization of full thickness skin wounds. Methods: In this study, mesenchymal stem cells were derived from rat adipose tissues and characterized by flowcytometry. Staining methods were used to evaluate their differentiatio...
